Gonal F - General Information
Human follicle stimulating hormone (FSH) preparation of recombinant DNA origin, which consists of two non-covalently linked, non-identical glycoproteins designated as the alpha- and beta- subunits. The alpha- and beta- subunits have 92 and 111 amino acids.
Pharmacology of Gonal F
Used for the treatment of female infertility, Gonal F or follicle stimulating hormone (FSH) stimulates ovarian follicular growth in women who do not have primary ovarian failure. FSH, the active component of Gonal F is the primary hormone responsible for follicular recruitment and development.
Gonal F for patients
Prior to therapy with Follistim�, patients should be informed of the duration of treatment and monitoring procedures that will be required. The risks of Ovarian Hyperstimulation Syndrome and multiple births, and other possible adverse reactions should be discussed.

Gonal F Contraindications
Follistim� (follitropin beta for injection) is contraindicated in women who exhibit:
1. Prior hypersensitivity to recombinant hFSH products.
2. A high circulating FSH level indicating primary ovarian failure.
3. Uncontrolled thyroid or adrenal dysfunction.
4. Tumor of the ovary, breast, uterus, hypothalamus or pituitary gland.
5. Pregnancy.
6. Heavy or irregular vaginal bleeding of undetermined origin.
7. Ovarian cysts or enlargement not due to polycystic ovary disease (PCOD).

Mechanism Of Action: FSH binds to the follicle stimulating hormone receptor which is a G-coupled transmembrane receptor. Binding of the FSH to its receptor seems to induce phosphorylation and activation of the PI3K (Phosphatidylinositol-3-kinase) and Akt signaling pathway, which is known to regulate many other metabolic and related survival/maturation functions in cells.
